Evaluating the effectiveness of the management of mechanisms nephropathy with prolonged use of glucocorticoids

Abstract: 

The aim of this work was to study free radical mechanism of nephropathy and to assess the capabilities of managing structural and functional impairment in the kidneys of rats with glucocorticoid-induced oxidative stress by the antioxidant "Thiophane".
The content and localization of proapoptotic protein factor Apaf-1 in the kidney parenchyma was identified by indirect immunohistochemical analysis carried out by the standard method. Blocking endogenous peroxidase was performed with 3% solution of hydrogen peroxide in the dewaxed sections. Specific monoclonal antibodies to a protein APAF-1 (LabVision, 1: 100) were used as primary antibodies. The avidin-biotin complex (UltraVHRPpolymerKIT, LabVision) was used for labelling secondary antibodies.
It is shown that the renal insufficiency in the animals receiving long-term glucocorticoid, is caused by damaging filtration apparatus components in the kidney glomerulus and by tubulointerstitial impairment. It is found that the prolonged use of glucocorticoids leads to increased expression of proapoptotic factors APAF-1 and lethal damage of nephrothelium by the apoptosis mechanism. Restriction of free radical processes through the use of polyfunctional sulfur-containing antioxidant "Thiophane" reduces the development of structural and functional disorders in the renal glomeruli and the number of epithelial cells with signs of apoptosis in the tubules of the nephrons.
